In the present work the characterization of different strains of Dunaliella salina from established cell culture collections and various isolates from solar saltworks located in the Canary Island, as well as one mutant, was carried out in order to assess the existence of intraspecific differences and to determine the potential productivity of each one. Morphological characteristics such as cellular size and cellular complexity and parameters linked to cellular physiology, such as pigment content or cellular growth rate, were determined by means of traditional techniques as well as flow cytometry. Efeito do meio Erd Schreiber no cultivo das microalgas Dunaliella salina, Tetraselmis chuii e Isochrysis galbana - DOI: 10.4025/actascibiolsci.v28i2.1037 Biological Sciences

As microalgas são utilizadas como fonte de alimento em aqüicultura. Neste trabalho cultivaram-se D. salina, T. chuii e I. galbana. O objetivo do trabalho consistiu em determinar o efeito do meio Erd Schreiber sobre o seu crescimento. Iniciou-se o cultivo com a mistura de 200 mg de Na2HPO4,7H2O, 100 mg de NaNO3 e 50 mL de extrato de solo. No monitoramento, manteve-se a temperatura entre 24 - 28oC, a salinidade a 34 ppt, à iluminação constante, a densidade celular com uma câmara de Neubauer e um microscópio binocular modelo ZEISS. Como resultado, I. galbana, D. salina e T. chuii atingiram 969 104 cel/mL, 457 x 104 cel/mL e 258,66 x 104 cel/mL, respectivamente, e os coeficientes angulares b foram 3,76 x 104 cel./mL/dia, 6,84 x 104 cel./mL/dia e 2,08 x 104...

Effects of n-propyl gallate on photosynthesis and physiological parameters in Dunaliella salina are affected by stressful conditions Braz. J. Plant Physiol.

We studied the effects of n-propyl gallate, which is a plastid terminal oxidase inhibitor involved in chlororespiration, on photosynthesis and physiological parameters in Dunaliella salina grown under different salinities and under low or high irradiance using chlorophyll a fluorescence transient measurements and pigment analysis. The inhibitor up to 1 mM had an additive significant effect on the photosynthetic efficiency in the cell suspensions grown under low salinity and irradiance. However, in the presence of high n-propyl gallate concentration (4 mM), there was a negative effect on all physiological aspects. In contrast, this high concentration of the inhibitor could enhance efficiency of electron transport and growth parameters under high irradiance....

Genetic polymorphism in eight Chilean strains of the carotenogenic microalga Dunaliella salina Teodoresco (Chlorophyta) Biol. Res.

Eight Chilean strains of Dunaliella salina obtained within a restricted geographic range, but exhibiting a high variability in their morphology, rate of growth and carotenogenic capacity, were analyzed by Random Amplified Polymorphic DNA (RAPD-PCR). Twenty of the 50 random primers (D, P, OPA and OPD series) that were tested amplified reproducible bands and were useful for comparative analysis of the strains. Of 107 polymorphic genetic markers, 49 were strain-specific. A great genetic variability was found among the strains in spite of their geographic proximity. In addition, phenetic analysis of the data showed close agreement between the morpho-physiological attributes and the genetic diversity of the strains

Hydrodynamics in a high-rate production reactor for microalgae cultivation affects the light history perceived by cells. The interplay between cell movement and medium turbidity leads to a complex light pattern, whose forcing effects on photosynthesis and photoacclimation dynamics are nontrivial. Hydrodynamics of high density algal ponds mixed by a paddle wheel has been studied recently, although the focus has never been on describing its impact on photosynthetic growth efficiency. Metabolic responses and β-carotene production by the unicellular green alga Dunaliella salina exposed to leaf extracts Acta Botanica

ABSTRACT The present work investigated the effects of aqueous extracts of eucalyptus ( Eucalyptus globulus) and elderberry ( Sambucus ebulus) leaves on β-carotene productivity in Dunaliella salina, a green microalga. Leaf extracts from eucalyptus have greater amounts of phenolics and flavonoids, as well as greater ferric reducing antioxidant potential than elderberry. The extracts of both species greatly inhibited growth of algal suspensions. However, chlorophyll and β-carotene concentration increased in cells treated with leaf extracts, and the highest values were detected in 1 % eucalyptus and 2 % elderberry extracts. Abstract Dunaliella salina, a single-celled marine alga with extreme salt tolerance, is an important model organism for studying fundamental extremophile survival mechanisms and their potential practical applications. In this study, two-dimensional differential in-gel electrophoresis (2D-DIGE) was used to investigate the expression of halotolerant proteins under high (3 M NaCl) and low (0.75 M NaCl) salt concentrations. Изучена жизнеспособность Dunaliella salina в зависимости от условий обезвоживания и длительности хранения в состоянии ангидробиоза Показано, что неинцистированные клетки D. salina сохраняют жизнеспособность в обезвоженном состоянии в течение 7 суток. Более длительное их хранение приводит к необратимым изменениям в клетках и потере способности восстанавливать жизнеспособность. Биохимические исследования зарегистрировали деструкцию пигментного ком- плекса. Зависимости инцистирования клеток от температуры обезвоживания, длительности дегидратации, остаточной влажности культуры после высушивания и времени хранения не выявлено. The dynamics of cell density of batch and semicontinuous culture of Dunaliella salina on different mediums were investigated. At the two stage productivity characteristics D. salina on biomass were estimated. It is shown that the productivity of the biomass of D. salina on the Trenkenshu medium is higher than on the Ben-Amotz medium in all stages of cultivation.

Продукционные характеристики роста и биосинтеза квазинепрерывной культуры зелёной микроводоросли Dunaliella salina Teod. IBSS Repository

Исследована динамика плотности, содержания хлорофилла а и белка квазинепрерывной культуры Dunaliella salina. На этапе накопительной культуры рассчитаны продукционные характеристики дуналиеллы по биомассе, хлорофиллу а и белку, с помощью которых построены прогностические кривые относительного содержания хлорофилла а и белка. Показано, что для получения максимальной продуктивности по биомассе клеток, белку и фотосинтетическим пигментам в квазинепрерывной культуре D. salina необходимо осуществлять обмен среды около 30 % от объёма. http://biotex.ibss.org.ua/
